It can be hard to see the filament glow in some preamp tubes but if it is working properly, the tube should feel warm after a minute or two. If not, and you know the wiring is correct and you didn't miss the jumper between pins 4 and 5, it will either be a bad solder joint or a broken wire causing the problem. I stopped twisting my heater wires because it's very easy to break the wire inside if you twist too much.

Just to keep you updated I’m more than half way done in gathering the best of the best to build a tweedle dee 5e3 this one should really have kick .

Don't go overboard buying expensive parts. I have tried all of the expensive stuff in builds and it never sounds any better than when I use standard quality parts. Don't forget that Leo Fender and Jim Marshall were buying the cheapest stuff they could find back then and their amps have stood the test of time. A well planned layout, clean lead dress and solid solder joints will have the biggest effect on the amp's performance.
